CAR-NK Microrobots with Programmable Swarm Navigation Hit Therapeutic Scale in Vivo
CAR-NK cells loaded with magnetic nanoparticles have been turned into biohybrid microrobots that can be magnetically steered to solid tumors at therapeutic doses in mice and nonhuman primates, according to a study in Science Advances. Programmable magnetic fields switch the cell swarms between two modes: a vortex mode for long-distance targeted transport and a ribbon mode for broad dispersion across the target site. In a metastatic leptomeningeal carcinomatosis model, intrathecal injection followed by magnetic guidance up the spinal canal delivered the microrobots into the brain's cerebrospinal fluid space, with real-time angiographic imaging confirming navigation in a nonhuman primate. The work points toward magnetically directed cell therapy for tumors that standard infusion routes struggle to reach. Elsewhere, the day's coverage includes a reinforcement learning anesthesia system matching anesthesiologists on hypoxemia in a 418-patient endoscopy trial, and China's first bulk actinium-225 production license issued in Chongqing.

Mitochondrial ATP promotes CD8+ T cell effector differentiation via phosphocreatine and BAF chromatin remodeling
Craig B. Thompson's team at Memorial Sloan Kettering Cancer Center reports in Cell that D-alpha-hydroxybutyrate (DAHB) increases mitochondrial ATP production by about 82% through fatty acid oxidation and distributes high-energy phosphate to the perinuclear region via the phosphocreatine-creatine kinase system. This supports BAF/mSWI-SNF-dependent chromatin remodeling, keeping effector gene loci such as Prf1, Ifng and Tnf open and thereby enhancing CD8+ T cell effector differentiation.
Science: Small intestinal epithelium sets differentiation by Hes1 oscillation frequency
On October 15, 2026, Katharina F. Sonnen's team at Utrecht University published a study in Science showing that intestinal epithelium determines cell differentiation direction through Hes1 oscillation frequency rather than average Notch levels.
CRISPR screens identify targets to rescue age-related T cell dysfunction in cancer.
Single-cell CRISPR screens identified Dusp5 and Zfp219 as drivers of CD8+ T cell dysfunction in aged tumors. Deleting Zfp219 caused epigenetic reprogramming, raised cytotoxic molecule expression, and combined with immune checkpoint inhibitors to clear tumors in aged mice; human ZNF219 levels in intratumoral CD8+ T cells correlated with worse survival after immunotherapy in older patients. The work names two targetable genes for rejuvenating antitumor immunity in aging.
Guangzhou Laboratory Finds SOX2 Condensates Drive Chemo Resistance in Lung Squamous Cancer, Designs Peptide Hx1R8
Hongjie Yao's group at Guangzhou National Laboratory reports in Signal Transduction and Targeted Therapy that a prion-like domain (PrLD) in SOX2 drives liquid-liquid phase separation to form condensates. These condensates directly bind and sequester chemotherapy drugs and weaken the DNA damage response, producing broad chemotherapeutic resistance in lung squamous cell carcinoma (LSCC).

Li Feng Team Shows PAK4 Kinase Promotes Exosome Release to Deepen Melanoma Immunosuppression
A team led by Feng Li at China Medical University, with Ruizhi Zhou as first author, reports in PNAS that PAK4 kinase intensifies melanoma immunosuppression through a dual mechanism that boosts exosome secretion. It phosphorylates ANXA2 at Ser26 in a kinase activity-dependent manner to promote multivesicular body docking, and retains TRIM21 in the nucleus independently of kinase activity, stabilizing SYT1 to support SNARE complex assembly and membrane fusion.
Muscle stem cells produce a protective Fibrillin-1 matrix to prevent precocious activation.
Fibrillin-1 (FBN1), a Notch-regulated matrix glycoprotein made by muscle stem cells themselves, is required to keep those cells quiescent. Deleting Fbn1 in muscle stem cells causes dose-dependent quiescence defects, loss of cellular projections, depletion of the stem cell pool, and progressive muscle decline, while human Marfan syndrome biopsies show abnormally activated satellite cells. TGFβ blockade rescues the defects of FBN1-deficient muscle, linking excess TGFβ signaling to stem cell dysfunction in MFS myopathy.
Nature Communications Paper Documents In-Utero Tumor Transmission Between Identical Twin Fetuses
A Nature Communications study reports pathological and genomic findings in identical twin girls in whom a tumor spread from one fetus to the other in utero via their shared placenta; both infants died soon after birth. Researchers from the Wellcome Sanger Institute, Great Ormond Street Hospital and other teams performed whole-genome sequencing and laser microdissection on 45 samples from the twins and their placenta, confirming the tumor originated in fetus A and metastasized to fetus B. The study also reviews a leukemia risk of roughly 15% to 25% in monochorionic twins and offers recommendations for screening pre-leukemia cells.
Lab-designed molecule reduces amyloid beta, the protein linked to Alzheimer's disease, in mice
A synthetic peptide, P33, designed by a team at Spain's IBEC, reduced amyloid beta (Aβ) levels and plaque-covered area in the cortex and hippocampus by 35.6% after eight weeks of dosing in APP/PS1 Alzheimer's mice. The molecule selectively inhibited BACE1 enzyme activity (over 50% reduction in vitro) without affecting the structurally similar BACE2, and no damage to other organs or changes in inflammation markers were observed. The preclinical study was published in Bioorganic Chemistry.

Multiplex programming of mappable, molecularly tagged genomic deletions
OPTIMuS PRIME-Del uses paired prime editing to program precise deletions that simultaneously insert a T7 promoter and barcode, allowing direct validation and quantification of edits in each cell. In K562 cells, pegRNA pairs targeted roughly 1,200 intronic candidate cis-regulatory elements, with a secondary screen of the ~100 most editable ones. Coupled with single-cell RNA sequencing, the platform identified intronic enhancers whose deletion cut host gene expression by 20-30% and a candidate enhancer-gene interaction spanning 124 kb.
Benchmarking the AlphaGenome Variant Impact score against SpliceAI on post-release ClinVar classifications and experimental splicing assays
AlphaGenome's claimed gains over other genome-wide scores do not hold on ground truth excluded from its developers' analyses. On 87,666 post-release ClinVar rare single-nucleotide variants, AVI reached an intronic AUROC of 0.957, indistinguishable from SpliceAI (difference −0.005) and Pangolin (−0.006), and it trailed SpliceAI by 0.070 on deep intronic variants. It also ranked splice-altering variants below raw SpliceAI on six splicing assays and gave benign evidence to only 24% of benign variants, against 90% for SpliceAI.
