Novartis Licenses Abogen mRNA T Cell Engager for $575 Million Upfront

Novartis has secured global rights to ABO2203, an mRNA-encoded CD19-targeted T cell engager from Abogen Biosciences, paying $575 million upfront with up to $7.2 billion in milestones attached, plus an option on follow-on RNA projects. The bet lands two weeks after a Ruijin Hospital team and Abogen published an exploratory study of the molecule in refractory immune thrombocytopenia in Cell. Elsewhere, the day brought a null result that matters: in a NEJM trial of 963 emergency department patients in septic shock, early vasopressors with restricted fluids matched higher fluid volumes and later vasopressors — median 76 days alive and out of hospital at day 90 in both groups — though pulmonary edema was rarer with the vasopressor strategy (0.6% vs 5.0%). Merck's remigromig also posted non-inferior vision outcomes versus anti-VEGF therapy in pivotal DME trials at one year.

Vasopressors or Fluids in Early Septic Shock.

Early vasopressor therapy with restricted fluids did not improve outcomes over higher fluid volumes with later vasopressors in emergency department patients with septic shock. In 963 intention-to-treat patients, days alive and out of the hospital at day 90 were a median of 76 in both groups (difference, 0.0 days; 95% CI, -2.7 to 2.7; P = 1.00). Pulmonary edema was less frequent with the vasopressor strategy (0.6% vs. 5.0%; P<0.001).

Extracellular matrix-targeted TNC-scFv armoring enhances CAR T cell persistence and antitumor activity in solid tumors.

Armoring CAR T cells with a secreted tenascin-C-targeted single-chain variable fragment (TNC-scFv) enhanced cytotoxicity, persistence, and reduced exhaustion in vitro and in xenograft tumor models. Single-cell transcriptomics showed the approach reprograms the tumor microenvironment toward a CD8+ T cell-enriched, functionally active immune landscape, and humanized tumor models showed robust antitumor responses with minimal systemic toxicity. Combining the armoring with interleukin-2 further increased efficacy while mitigating cytokine-associated toxicity, establishing extracellular matrix-directed armoring as a generalizable strategy for solid tumors.

Biomarker and Bioorthogonality Dual-Locked Chemiluminescent Probes for Cancer Urine Test

Chemiluminescent tetrazine probes (CLTZs) stay dark until a bioorthogonal reaction with bicyclononyne (BCN) unlocks emission, enabling biomarker and bioorthogonality dual-locked cancer urine testing. Pairing an H2O2 and BCN dual-locked probe (HBCP) with a cancer-activatable urinary reporter (CBUR) gave in vivo tumor imaging with a signal-to-background ratio of 438 and detected lung metastasis in urinary assays with up to 23.9-fold signal enhancement over healthy mice. The work adds a molecular design route to noninvasive chemiluminescent diagnostics.

Epigenetic Remodeling of Hippocampal CA2 Perineuronal Nets Underlies Sevoflurane Neurotoxicity in the Developing Brain

A team led by Zhang Hui, Zhang Haopeng, Liang Lirong and Wu Shengxi reports in Advanced Science that sevoflurane exposure during the developmental window injures the brain through epigenetic regulation that remodels perineuronal nets in hippocampal region CA2. The work ties repeated sevoflurane exposure in children, a risk previously linked to long-term learning and memory impairment, to a defined molecular mechanism acting on CA2 excitatory pyramidal neurons, the cells that carry social memory. It identifies a cellular and epigenetic target for the neurodevelopmental risk of the most widely used inhaled anesthetic in infants and young children.

Scalable methylome-transcriptome profiling resolves clonal organization in human hematopoiesis

Droplet-MT jointly profiles transcriptomes and methylomes at ~10,000 cells per library, using methylation epimutations for mutation-independent clonal reconstruction and label transfer at ~0.2% CpG coverage to cut sequencing cost. Applied to ~21,000 paired profiles from healthy blood and longitudinal acute myeloid leukemia, it resolved small and expanded clones with heterogeneous lineage output and distinguished malignant clonal abundance changes, state remodeling within persistent clones, and recovery of healthy polyclonal hematopoiesis. Malignant monocytes stayed transcriptionally and epigenetically distinct from healthy and recovery-associated cells despite treatment-associated remodeling.

Disrupted chromatin architecture blocks B lymphopoiesis in Down syndrome

Three-dimensional genome architecture organizes human fetal haematopoiesis and explains the B-lineage defect in Down syndrome. Using ChromE-seq, a new single-cell assay jointly profiling chromatin conformation and transcription, the authors built a multimodal atlas of ~400,000 haematopoietic cells from disomic and trisomy 21 fetal liver. Trisomy 21 disproportionately disrupted B-lineage architecture, weakened an EBF1-centred regulatory circuit, and deleting a T21-sensitive EBF1 loop anchor reproduced the CLP-to-B-progenitor bottleneck seen in T21.

An iPSC-based experimental framework for investigating phenotypic variation associated with Jomon ancestry in the present-day Japanese population

Jomon ancestry, measured by the Jomon allele score, tracked with renal-related traits and serum triglycerides in 34,152 Tohoku Medical Megabank participants, and donors at JOM extremes (n = 10) showed group-level differences in immune-related gene expression. But when hiPSCs from those donors were differentiated into hepatocyte-like cells, intracellular triglycerides and lipid-related gene expression did not differ between groups. The work establishes a genetically characterized hiPSC framework for testing ancestry-associated phenotypes at the cellular level.

Ayahuasca plant genomes reveal retrotransposon-derived centromeres and contrasting routes to DMT and β-carbolines

Genome assemblies of the two ayahuasca source plants reveal contrasting histories: Psychotria viridis is an autopolyploid hexaploid (2n = 6x = 66) while Banisteriopsis caapi is diploid (2n = 2x = 20), yet centromeres of both are enriched in CRM-type Ty3/Gypsy retrotransposons. In P. viridis a single multifunctional methyltransferase catalyzes both N-methylations of tryptamine to DMT, whereas B. caapi makes beta-carbolines via dedicated enzymes plus a non-enzymatic, acetaldehyde-dependent Pictet-Spengler cyclization followed by enantioselective oxidation to harmaline. The work supplies the genetic structure of traditionally recognized cultivated varieties and the enzymes behind the alkaloids combined in the decoction.

PreGame Algorithm Identifies Broad-Spectrum Anti-Tumor Gamma-Delta T Cells in Multiple Myeloma

A machine learning algorithm named PreGame, published in Nature, identifies broad-spectrum anti-tumor gamma-delta T cells in multiple myeloma. The work addresses a long-standing question of whether tumor-infiltrating gamma-delta T cells recognize myeloma cells directly through their TCR or via receptors such as NKG2D and NKp30 binding ligands on cancer cells. PreGame offers a computational route to select these cells, which combine innate-like and adaptive-like properties, for potential cell therapy.

Structural variants from donor-specific assemblies affect recurrent chromatin contact divergence in humans

Donor-specific genome assemblies markedly improve mapping of haplotype-resolved chromatin contacts: across 354 human and 12 nonhuman ape haplotypes, 13.2% of cis Hi-C contacts showed >25 kbp changes in inferred genomic separation and 2.8% more valid contact pairs were recovered compared with standard references. Haplotype-specific chromatin differences are selective, concentrating in 1,790 recurrent contact difference hotspots linked to 2,495 chromatin-structure-associated structural variants enriched in regulatory contexts and associated with expression variation; over 90% of these SVs arose in the human lineage. The work argues for building pangenome haplotype diversity into studies of 3D genome organization.

HealthFlux world model keeps 20-year health predictions accurate and simulates drug effects across 1,256 diseases

HealthFlux, a pan-modal human health world model posted on medRxiv by Stanford postdoc Ying Kejun and colleagues, maintains a continuous 384-value health state trained on UK Biobank data spanning 11 data types including records, blood tests, plasma proteomics, metabolomics and MRI. In about 60,000 held-out participants it predicted 1,010 diseases and death over 5 years with an average AUROC of 0.832 versus 0.805 for RisQ, and after 20 years of autonomous rollout without new data it held 0.837 while Delphi-2M fell to 0.576. Simulated drug effects for 210 drug-disease combinations matched published clinical trials within 18%, and screening 259 drugs across 1,256 diseases suggested metformin cut 10-year mortality risk by about 21% and simvastatin cut major disease risk by about 35%.
