Two B7-H3 ADCs Post Identical 0.46 Survival Hazard Ratios in Relapsed Small Cell Lung Cancer

Two B7-H3 antibody-drug conjugates delivered Phase III overall survival wins in relapsed small cell lung cancer at the September 2026 WCLC presidential forum in Seoul. Tambotatug pelitecan (YL-201, MediLink Therapeutics) in TAISHAN-302 and risvutatug rezetecan (HS-20093, Hansoh/GSK) in ARTEMIS-008 each hit an overall survival hazard ratio of 0.46, rewriting a second-line standard that had held for three decades and putting B7-H3 ADCs in direct contention with DLL3 bispecifics in the same setting. The day also brought a negative Phase 3 for nicotinamide riboside in early Parkinson disease (MDS-UPDRS worsening of 2.72 points vs placebo, P = .02), FDA approval of pritelivir for acyclovir-resistant herpes lesions, and Kodiak's dual Phase III DAYBREAK success in ophthalmology.

Cervical cancer trial finds similar early recovery results after robot-assisted versus open surgery

Robot-assisted radical hysterectomy for early-stage cervical cancer produced fewer intraoperative complications than open surgery (5.7% vs 10.0%), but the difference mainly involved less serious events. In the randomized RACC study of 900 patients across European hospitals, complications within 30 days were nearly identical (33.4% vs 34.8%), and overall early recovery and most quality-of-life measures were similar. Recurrence-free survival, the primary outcome, is still being followed.

DNA Typewriter Reconstructs Mouse Cell Lineage Tree from Zygote to E13.5

A DNA Typewriter recorded cell lineage in a living mouse, and from a single embryo the team reconstructed a phylogenetic tree rooted at the zygote, time-calibrated to E13.5, containing 1.28 million cells annotated with transcriptomes. The work, by Chengxiang Qiu with Jay Shendure's group at the University of Washington, published in Science on October 9, 2026, closes a 50-year gap since Sulston manually traced a nematode's cell divisions. The tree reveals clonal dominance during gastrulation, hierarchical structure coupled to cell type, and ancestral state paths.

Message in a bottleneck: Nested founder effects from French Polynesia to Rapa Nui and Hawai'i.

Polynesians experienced founder effects an order of magnitude stronger than those described in European-descent groups such as the Finnish and Amish during their settlement of Eastern Polynesia. Whole-genome sequences spanning Polynesia show that Hawai'i and Rapa Nui, the two most bottlenecked populations, were likely settled by the same ultravoyagers with ancestral roots in Mangareva. The analysis identifies a loss-of-function FAN1 mutation common in Eastern Polynesia (>1%) but absent from gnomAD, offering a target for population-specific genetic testing.

Ancient viral DNA helps embryonic stem cells grow—and cancer hijacks the same switch

A human endogenous retrovirus, HERVH, drives production of the calcium-buffering protein calbindin in embryonic stem cells, and the same pathway had earlier been seen helping cancer cells survive by preventing them from aging. Editing human embryonic stem cells to remove the HERVH switch left cells unhealthy, failing to grow or specializing prematurely, and blastoids and post-gastrulation amnioids lacking HERVH-driven calbindin could not form properly. Primate genome comparisons place the HERVH integration after the split from orangutans, roughly 13 million years ago.

Distinct malaria parasites may help explain sickle cell trait's protection against disease

People with the sickle cell variant are infected by genetically distinct malaria parasites and are a tenth as likely to develop symptomatic disease, according to sequencing of more than 2,000 people in Mfou, Cameroon, and the parasites infecting them. Asymptomatic infections affected three-quarters of the healthy study population, and parasite varieties differed by sickle cell status. The finding shifts explanation of sickle cell trait's protection toward the parasite genotype, not only the host response.

Electrical ablation boosts CAR-NK cell therapy against liver cancer, offering new strategy for refractory solid tumors

Irreversible electroporation (IRE) combined with GPC3-targeted CAR-NK cells, made with lipid nanoparticle gene delivery, produced substantially stronger tumor control than either treatment alone in hepatocellular carcinoma. IRE reshaped the immunosuppressive tumor microenvironment: chemokine levels rose, recruiting NK cells early, while IRE-induced reactive oxygen species left residual tumor cells more susceptible to NK cell killing. Across multiple preclinical models the combination showed superior tumor suppression and prolonged survival.

Oligo(uridine) tailing and exonucleolytic decay drive 40S ribosome degradation in response to stress

Stress-induced degradation of 40S ribosomes proceeds through 3′ oligo(U) tailing of 18S rRNA. TUT7 is recruited to ubiquitylated 40S ribosomes and uridylates the 18S rRNA, and iterative uridylation followed by 3′-5′ decay by the exonuclease DIS3L2 drives degradation of the 18S rRNA and the 40S ribosome. The work identifies the enzymatic route linking ribosome ubiquitylation to decay.

The Red Blood Cell Surface Defines a Distinct Tumor DNA Compartment for Liquid Biopsy

Red blood cells tether and retain tumor-derived DNA on their surface, forming a distinct circulating pool of RBC-bound tumor DNA (rtDNA), with desialylation increasing capture in a Toll-like receptor 9-dependent manner. In mice bearing lung cancer xenografts, rtDNA was detected in all animals and tracked tumor burden while plasma tumor DNA was undetectable; in patients with lung cancer, most detected mutant tumor DNA was RBC-associated and concordant with matched tumor tissue. The findings challenge the plasma-centric liquid biopsy paradigm by identifying the RBC surface as a tunable reservoir of tumor DNA.

Scientists identify the neuroblastoma cells that signal poor outcomes

A gene expression signature distinguishes malignant mesenchymal neuroblastoma cells, the population linked to high-risk disease and treatment resistance, from healthy mesenchymal cells that had confounded earlier efforts. Built from 54 tumors across 50 patients using single-cell RNA sequencing and spatial omics, the signature improved outcome predictions in an independent tumor database, where a signature from decades-old cell lines did not. Patient-derived xenografts, organoids and new cell lines capturing both cell states are available through the St. Jude Childhood Solid Tumor Network.

Current age-based genetic testing in patients with cancer may miss the majority of inherited gene variants

Universal germline testing of 39,184 patients with solid tumors found germline pathogenic variants in 16.3% overall, and restricting testing to patients under age 50 would have excluded 4,601 carriers, or 72% of all patients with pathogenic variants. Even with cancer type-specific onset definitions, pathogenic variants appeared in 12.3% of late-onset cancers. The findings support germline genetic testing for every patient with cancer regardless of age at diagnosis.

A mitotic restriction point for extrachromosomal DNA hubs in cancer cells

Mitosis is the critical window in which extrachromosomal DNA (ecDNA) hubs are organised and inherited across cell generations. An optical screen identified the mitotic-entry regulator WEE1 as a key regulator of hub integrity and oncogene expression, with CDK1- and CDK2-regulated programmes cooperating to carry unresolved damage into mitosis and reorganise ecDNA tethered to mitotic chromosomes. Live-cell imaging shows hub architecture is remodelled during mitosis and maintained through interphase, a coupling conserved across cancer types and ecDNA-associated oncogenes, defining a mitotic restriction point and a potential therapeutic window for ecDNA-driven cancers.

40S ribosome remodeling triggers 18S rRNA U-tailing and DIS3L2-dependent decay

RIOK3 remodels ubiquitinated 40S ribosomal subunits so that the 3′ end of 18S rRNA is exposed and uridylated, marking it for DIS3L2-mediated degradation. The finding couples ribosomal protein ubiquitination triggered by translational perturbations to selective 18S rRNA turnover, filling a gap in how 40S ribosome quality control is executed.

Random mtDNA mutagenesis maps genotype-phenotype associations across metabolic contexts and lineages

mtDIVE, a random C→T mutagenesis platform, enables pooled mitochondrial forward genetics in human epithelial cells and zebrafish embryos, generating molecules from single substitutions to more than 100 mutations. Selection on mutant genomes and their transcriptional consequences depend on metabolic state and developmental lineage: obligate OXPHOS eliminates highly mutated cells by apoptosis, while glycolytic conditions promote their competitive displacement. Individual loci have modest, non-redundant effects that predict cell state beyond mutation burden, and cancer-recurrent variants are enriched among mutations linked to reduced mitochondrial transcription.

Systematic mapping of enhancers, silencers, and topological elements with prime editing deletion screens

Swap-seq, a method combining pooled twin prime editing with flow-sorting, deletes hundreds of endogenous regulatory sequences and measures their quantitative effects on nearby gene expression. Applied to PPIF regulation in THP-1 monocytes, it quantifies distal and intronic enhancers, characterizes CTCF insulators, and uncovers a silencer that represses neighboring enhancers through HDAC-mediated mechanisms, with hundreds of genomic elements sharing its signatures. The resulting data show sequence-to-function models capture local element activity but fail to link elements to distal target genes.

James Kirkland presents senolytics and cell-clearing strategies at CSHL lecture

James Kirkland outlined senolytics, agents that selectively clear tissue-damaging senescent cells, in a Cold Spring Harbor Laboratory aging symposium talk. He traced the field's discovery logic to an analogy with the death-resistance of leukemia and lymphoma cells, which led to screening natural products and repurposed drugs that briefly switch off senescent cell defenses. He said cancer settings need a new class of senolytics or "senescent sensitizers" to remove senescent cells that may carry cancer-causing mutations, and that several agents have entered clinical trials.
