Li Feng Team Shows PAK4 Kinase Promotes Exosome Release to Deepen Melanoma Immunosuppression

In brief

A team led by Feng Li at China Medical University, with Ruizhi Zhou as first author, reports in PNAS that PAK4 kinase intensifies melanoma immunosuppression through a dual mechanism that boosts exosome secretion. It phosphorylates ANXA2 at Ser26 in a kinase activity-dependent manner to promote multivesicular body docking, and retains TRIM21 in the nucleus independently of kinase activity, stabilizing SYT1 to support SNARE complex assembly and membrane fusion.

Why it matters

The paper defines kinase-dependent and kinase-independent mechanisms by which PAK4 drives exosome release, supporting combining PAK4 inhibitors with immune checkpoint blockade.
