Intraoperative Dobutamine Plus Norepinephrine to Prevent Postoperative Acute Kidney Injury in Septic Shock: A Randomized Clinical Trial.

Intraoperative dobutamine (up to 5 µg/kg/min) plus norepinephrine reduced postoperative acute kidney injury within 7 days in adults with septic shock undergoing surgical source control: AKI occurred in 22.8% (66/290) with the combination vs 34.7% (102/294) with norepinephrine alone (adjusted risk ratio 0.64 [95% CI, 0.49-0.83]). Stage 2 and 3 AKI were also lower, though tachycardia was more frequent. The trial, conducted in 39 tertiary hospitals in China, provides randomized evidence for a guideline recommendation previously based on very low-certainty evidence.

Newer Migraine Drugs Linked to a 40% Higher Risk of Miscarriage in Pregnant Women, Study Suggests — Study Raises Questions About the Pregnancy Safety of CGRP-Targeting Treatments, but Further Research Is Needed to Determine Whether the Medications Directly Contribute to Pregnancy Loss.

CGRP-targeting migraine drugs were associated with a higher miscarriage rate in the AM-PREGNANT cohort released at AHS 2026: roughly 5% in users versus about 2% in non-users, a relative increase of about 45%. The observational cohort covered 7,119 women and 7,579 pregnancies and cannot establish that the drugs cause pregnancy loss. Clinicians prescribing CGRP agents to women of childbearing potential will need to weigh this signal against the drugs' benefits.

Milk osteopontin alters the infant microbiome to drive DC hematopoiesis and disease tolerance.

Milk-derived osteopontin protects neonatal mice against severe lower respiratory infections by supporting dendritic cell hematopoiesis in the developing liver and lung. Mice lacking milk OPN were predisposed to viral and bacterial sLRI; oral OPN raised Lactobacillaceae and serum 3-phenyllactic acid, a PPARγ agonist, and PLA or rosiglitazone restored lung DC hematopoiesis through CCL25-mediated progenitor recruitment. Depleting plasmacytoid DCs, neutralizing stem cell factor, or deleting airway epithelial Flt3L attenuated the effect.

Functional diversification of the cephalopod proteome by RNA editing.

Proteome-scale analysis of the squid Doryteuthis pealeii confirms translation of 7,997 RNA-recoded sites and shows how recoding alters protein stability, localization, post-translational modifications and enzymatic activity. Recoding of the E3 ligase MARCHF5 shifts substrate ubiquitylation and perturbs mitochondrial homeostasis, linking cephalopod RNA editing to organelle function.

SMARCA4 Confirmed as Tumor Suppressor in Germinal Center B Cell Lymphoma Mouse Models

A researcher trained at MD Anderson under M. Green and now at the Chinese Academy of Medical Sciences/CIMR presented work establishing SMARCA4 (BRG1), the shared ATPase of the SWI/SNF chromatin remodeling complex, as a tumor suppressor in germinal center-derived B cell lymphoma. In mouse conditional knockout models, heterozygous SMARCA4 loss skewed germinal centers toward the dark zone and expanded recycling centrocyte-to-centroblast cells, while homozygous loss sharply reduced germinal centers; in the Eμ-Myc lymphoma model, heterozygous loss shortened survival. The SWI/SNF inhibitor FHD-286 reduced tumor burden and extended survival in mutant tumors, with mechanistic work showing loss of accessibility at transcription factor motifs including IRF4, BCL6 and PAX5 targets.

A Compact Regulatory Logic Governs Fibroblast Cell State Plasticity in Fibrosis

A compact regulatory logic controls fibroblast activation in fibrosis: repressing 280 conserved disease-activated enhancers via CROP-seq in ~550,000 fibroblasts mapped 15 gene programs governing the profibrotic state, converging on MEOX1 as a central regulator. Fibroblast-specific Meox1 deletion in mice improved cardiac function and reduced fibrosis in heart failure. Repressing just six enhancers with multiplexed dCas12a-KRAB collapsed the profibrotic network and attenuated macrophage inflammation.

Whole-Body HOListic Imaging in Larval Zebrafish Maps Cross-Organ Cell Communication

HHMI Janelia's HOListic system performs whole-body in vivo functional imaging in larval zebrafish ubiquitously expressing GCaMP, extracting roughly 400 activity clusters that map largely to single organs and often to subcompartments. Applying it revealed unreported cold sensitivity in chondrocytes, a hindbrain-gut topographic mapping confirmed by optogenetics, and a possible brain control mechanism in which hypoxia shifts blood flow away from viscera and brain inhibition restores flow within seconds. The work argues that broad measurement across context can find control nodes that the classic perturb-and-draw-arrows approach misses.

Petter Brodin Lecture Maps Environmental and Hormonal Drivers of Human Immune Variation

Petter Brodin's Global Immunotalks lecture covered human immune system variation and development. He described fixing whole blood at collection to retain neutrophils and lower technical CV, twin studies from Stanford and Mark Davis showing non-heritable environmental factors dominate most immune measures and vaccine responses, and a Karolinska birth cohort of over 500 children linking breast milk oligosaccharides, bifidobacteria and tryptophan metabolites such as indolelactic acid to reduced early gut inflammation. He also presented data on 23 transgender men given testosterone, in which type I interferon pathways were suppressed and TNF/NF-kappaB inflammation induced.

David Julius Used Capsaicin and Menthol Probes to Identify the TRPV1 and TRPM8 Temperature Sensors

David Julius identified the capsaicin receptor TRPV1, the first cloned vanilloid receptor, using capsaicin as a probe with cDNA library screening and calcium imaging, and confirmed with patch clamp that it is an ion channel mediating Na+/Ca2+ influx that also responds to heat at the human threshold between warmth and burning pain. Using menthol as a probe, he then identified the TRPM8 cold receptor, which opens below about 26 degrees Celsius; TRPM8 knockout mice could not distinguish 20 degree from 30 degree platforms, validating TRP channels as key molecular detectors of environmental temperature. These channels are now targets for developing new analgesics.

Rusty Gage Links LINE-1 Retrotransposon Activity to Neuronal Aging and Alzheimer's Disease

Rusty Gage described how LINE-1 retrotransposons, about 40% of the human genome, are silenced in mature neurons as RNA condensates that dissolve with aging, freeing LINE-1 RNA to trigger cGAS-STING inflammation and neuronal senescence. His team derived neurons from patient fibroblasts in about three weeks, preserving donor age, with a skin biopsy bank from 150 healthy elderly and Alzheimer's and mild cognitive impairment donors. Reverse transcriptase inhibitors such as 3TC reduced inflammation in AD-derived neurons in vitro, mouse model tests are under way, and Gage expects AD-related human trials within about two years.

Michael Hall Explains How TOR Controls Cell Growth, From Yeast Rapamycin Mutants to Human Disease

Michael Hall explains that TOR, a highly conserved protein kinase first identified in 1991 through yeast mutants resistant to rapamycin, controls cell growth by size and mass rather than cell-cycle G1 progression. Rapamycin exerts toxicity as a complex with FKBP12 binding specific sites on TOR. TORC1 and TORC2 are conserved from yeast to humans, and TOR dysregulation links to cancer, cardiac hypertrophy, obesity and type 2 diabetes, with metformin partly acting through TOR inhibition.

Fc receptor engagement lets anti-CD25 antibodies deplete intratumoral Tregs, experts explain

Experts on ImmuneZoom discuss how anti-CD25 antibodies deplete intratumoral Tregs through Fc receptor mechanisms. A non-IL-2-blocking CD25 antibody (NIB) achieved 100% complete regression as a single agent in inflamed tumors, and in a glioblastoma model, combined with an anti-EGFRv3 antibody, also reached 100% complete regression via IL-2-driven CD8 expansion and IFN-gamma activation of macrophages. The humanized anti-CD25 RG6292 depleted Tregs in blood and was well tolerated but achieved under 2-fold, unstable intratumoral depletion, which may explain clinical failure; the team now studies boosting Fc receptor activity in the tumor microenvironment.

The Next Generation of Weight-Loss Drugs Is Almost Here

A Reddit thread on r/GLP1ResearchTalk debates next-generation amylin-based weight-loss drugs presented at EASD 2026, where CagriSema (cagrilintide plus semaglutide) showed 22.4% weight loss over 52 weeks versus placebo (p<0.0001) and Lilly reported Phase IIb data for its amylin analogue eloralintide. Commenters split over whether high dropout rates in higher-dose eloralintide plus tirzepatide combinations signal tolerability problems, with others blaming faster titration; Lilly has not disclosed dropout reasons. Novo Nordisk submitted CagriSema's NDA to the FDA in December 2025, with a decision expected in Q4 2026.

Thymic innate-like T cells shape CD8 tolerance via T-to-T antigen presentation, talk finds

An expert lecture on ImmuneZoom described how thymic innate-like T cells, including NKT cells, reside long-term in the thymus and express inflammatory mediators such as IFN-gamma, forming a reservoir of inflammation-associated self-antigens. Experiments with GFP/IFN-gamma model antigen, Jedi TCR transgenic mice and thymic organoids showed these cells induce negative selection of CD8 T cells through direct T-to-T antigen presentation across roughly a 700-fold affinity range; as few as 0.1–0.55% of thymic innate-like T cells efficiently eliminate self-reactive CD8 T cells. Unpublished work identified V-delta-7 TCR recognition of the IL-17 receptor A chain as a ligand driving gamma-delta T cell selection.

Shanghai Institute of Materia Medica's Bo Shan explains how metabolic signals shape immune cell phenotypes

Dr. Shan of the Shanghai Institute of Materia Medica gave a lecture on how metabolic signals regulate immune cell phenotypes, linking fat browning and liver regeneration. His team found that deleting the mitochondrial protein LUMP1 in adipocytes blocks cold-induced browning by degrading SDHB and stabilizing succinate, which alters the succinate/α-KG ratio and histone methylation. In liver regeneration, a CD36-positive monocyte-derived macrophage subpopulation takes up lipids, activating the IRE1-XBP1 pathway to raise IL-6 and drive hepatocyte proliferation, with IL-6 rescue restoring proliferation when CD36 or IRE1 is knocked out.

Danisco yogurt strain research showed CRISPR is a bacterial adaptive immune system

Rodolphe Barrangou of Danisco, a Danish food ingredients maker and leading producer of yogurt and cheese cultures, traced how industrial phage resistance work revealed CRISPR function. Archived frozen strains differed from their parents only by an added repeat and spacer in the CRISPR array, and adding a spacer conferred resistance while removing it or knocking out CAS9 abolished it. The team published its first high-impact CRISPR paper in March 2007, proving CRISPR is a bacterial adaptive immune system.

Selective connexin43 hemichannel-blocking peptide improves cognition in Alzheimer's mouse model

Researchers found that connexin43 hemichannels open abnormally on reactive astrocytes and microglia near amyloid plaques in Alzheimer's disease brains and APP/PS1 mice, mediating calcium influx and ATP/glutamate release that damage neurons. In collaboration with Aha, they used a TAT-Cx43 peptide, delivered in VPN lipid nanoparticles giving seven-day brain exposure after intraperitoneal injection, to selectively block hemichannels while sparing gap junctions. In APP/PS1 mice, weekly dosing either from 8 months for seven weeks or from 3.5 months for nine weeks improved cognition, reduced oxidative stress and dendritic spine loss, and shifted microglia toward a neuroprotective state.

Ali Shilatifard Links Transcription Elongation Control to Disease and Aging at CSHL Leading Strand Talk

Ali Shilatifard discussed transcription elongation regulation at a CSHL Leading Strand lecture, arguing on the basis of 30 years of his laboratory's work that elongation control is central to gene expression and that its disruption can cause disease. He connected single-point mutations in the super elongation complex to neurodevelopmental disorders such as Coffin-Lowry syndrome, and noted elongation factors are also linked to cancer. In a session on aging, he discussed whether elongation factors can be targeted inside cells to counter senescence-related phenotypes.

Ron Vale Recounts the Discovery of Kinesin, a Microtubule-Based Motor Protein

Ron Vale tells the story of how kinesin, a motor protein that moves organelles along microtubules, was discovered. Work at Woods Hole (MBL) used video-enhanced microscopy developed by Allen, Inoué and others to visualize axonal transport in squid axons directly for the first time. A control experiment meant to rule out interference from soluble protein fragments instead confirmed that organelles themselves move along microtubules, leading to the purification of kinesin and a change in Vale's own research trajectory.
