Ivonescimab Cuts Death Risk 27% Versus Keytruda in Frontline Lung Cancer

Akeso and Summit's ivonescimab cut the risk of death by 27% versus pembrolizumab in the phase 3 Harmoni-2 trial, testing the bispecific as first-line therapy for PD-L1-positive non-small cell lung cancer. The survival benefit was especially pronounced in patients with high PD-L1 expression, strengthening the case for a credible challenger to Keytruda in one of oncology's largest markets. The day also brought FDA action: accelerated approval of AstraZeneca's camizestrant plus CDK4/6 inhibitors for ESR1-mutant breast cancer, and clearance of Bayer's Hyrnuo for first-line HER2-mutant lung cancer, setting up a rivalry with Boehringer Ingelheim's Hernexeos.

Sintilimab‑related cystitis/ureteritis in high‑grade endometrial stromal sarcoma: A case report and structured literature review

Sintilimab can trigger cystitis/ureteritis that mimics infection, a 44-year-old woman with high-grade endometrial stromal sarcoma shows. After three cycles of sintilimab with nab-paclitaxel and carboplatin she developed severe urinary frequency, urgency and dysuria unresponsive to empirical antibiotics; methylprednisolone, with later piperacillin-tazobactam for cultured Enterococcus faecalis, resolved symptoms. Rechallenge caused only mild symptoms and a switch to tislelizumab brought no recurrence. The report pairs the case with a structured literature review for managing such urological irAEs.

Myeloperoxidase inhibition with mitiperstat in heart failure with preserved or mildly reduced ejection fraction: a randomized phase 2b trial

Mitiperstat, a myeloperoxidase inhibitor targeting the neutrophil–MPO inflammatory pathway, did not improve symptoms or exercise function in patients with heart failure with preserved or mildly reduced ejection fraction in a phase 2 randomized clinical trial. The finding argues against this anti-inflammatory approach as a symptomatic therapy in this population.

A binding ban, and its limits: Japan legislates on heritable human genome editing.

Japan enacted the Act on Regulation of Handling of Human Genome-Edited Embryos, etc. on 17 July 2026, promulgated a week later as Act No. 70 of 2026, criminalizing the reproductive use of genome-edited human embryos. The law replaces non-binding 2019 guidelines that barred transfer of edited embryos to a uterus without penalty, and closes a gap left by the 2000 Act on Regulation of Human Cloning Techniques, which did not cover genome-edited embryos. Tsutomu Sawai of Hiroshima University notes that a national criminal ban can still be evaded by seeking the procedure abroad.

STAT+: Leukemia drug data suggested a different way to treat diabetes. A new biotech will test that idea

Kura Oncology is spinning off its menin inhibitor programme into a new company, Caspian Therapeutics, which launches with $50 million in financing led by BVF Partners and joined by Eli Lilly. Animal studies of Kura's menin-targeting leukemia therapy showed the approach also helped control blood sugar, so Caspian will develop menin inhibitors for diabetes and other cardiometabolic diseases, taking its lead small molecule into an early-phase diabetes trial and backing a second candidate. Kura retains about 50% ownership, Kura CEO Troy Wilson becomes executive chair, and Robert Spencer was named to lead Caspian.

Cell-type signatures of Alzheimer's disease shared across population groups.

Cell-type signatures of Alzheimer's disease are shared across Latin, white and African American population groups. Single-nucleus RNA sequencing and ATAC-seq of post-mortem cortical and subcortical brain tissue identified disease-associated microglial, astrocytic and neuronal clusters in all three groups, plus continuous gene-expression factors in astrocytes and oligodendrocytes. Molecular programs defined six subgroups of individuals with cognitive impairment that span all three populations and are not captured by neuropathology.

Design of optimized epigenetic regulators for durable gene silencing with application to PCSK9 in nonhuman primates.

An optimized TALE-based epigenetic regulator, EpiReg-T, silenced PCSK9 in macaque liver with over 90% efficiency lasting 343 days after a single lipid nanoparticle dose, lowering LDL cholesterol. In mice, EpiReg-T reached 98% efficiency versus 64% for the initial dCas9-based design. Multiomic analyses showed minimal off-target effects in monkeys, mice and human-derived cells, and the platform can be retargeted to other genes by reengineering the DNA-binding domain.

Rb-driven transcription limits its tumour-suppressive effects in breast cancer.

CDK4/6 inhibition redistributes hypophosphorylated Rb to promoters and enhancers, where it not only represses cell cycle genes but also promotes expression of oestrogen-responsive genes by joining ER-rich transcriptional hubs, partly through KDM5A. This Rb-driven ER transcriptional program is pro-proliferative: anti-oestrogen therapy neutralizes it in endocrine-sensitive tumours, but it persists in ESR1-mutant breast cancer and limits CDK4/6 inhibitor efficacy. The work reframes Rb as a dual-function transcriptional regulator.

Zhongshan Hospital Team Maps Two Key Events Driving Liver Cancer Metastatic Colonization in Science

A multi-omics study led by Fan Jia's team at Zhongshan Hospital, published in Science, used Stereo-seq, single-cell and chromatin accessibility data to identify two key events in liver cancer metastatic colonization: disseminated tumor cells with high Phgdh expression establish an immune-desert niche in a quiescent state, and Cx3cr1-positive macrophages later drive metastatic reactivation. PHGDH-positive tumor cell features were cross-validated in public single-cell datasets from lung adenocarcinoma and ovarian cancer.

TM184C is a GPCR-like regulator of intercellular exchange and autophagy.

Mining millions of AlphaFold2 models revealed two human families of 'superdark' seven-transmembrane proteins, TM184 and PRRT, with structural homology to GPCRs and hallmark GPCR activities including β-arrestin recruitment and GRK-dependent phosphorylation. TM184C, the most broadly expressed and conserved member, localizes to intracellular vesicles that move along microtubules and promote tunnelling nanotube- and tumour microtubule-like intercellular connections mediating organelle sharing. TM184C also limits LC3B lipidation and autophagosome accumulation, and human TM184C restores autophagic body homeostasis in yeast lacking its homologue Hfl1.

Live imaging of late-stage preimplantation human embryos reveals de novo mitotic errors.

Light-sheet live imaging of late-stage preimplantation human embryos, enabled by mRNA electroporation of nuclear DNA labels, shows chromosome segregation errors arising de novo immediately before implantation, including multipolar spindles, lagging chromosomes, misalignment and mitotic slippage. Most lagging chromosomes are passively inherited rather than reincorporated, and most labeled cells remain externally positioned, consistent with placental fate. The findings question clinical uses of preimplantation genetic testing for aneuploidy.

Polyamines buffer labile iron to suppress ferroptosis.

Polyamines buffer redox-active iron in mammalian cells, linking polyamine metabolism to ferroptosis. A genome-wide CRISPR screen revealed a synthetic lethal dependency between polyamine depletion and the ferroptosis suppressor GPX4, and polyamine deficiency redistributed cellular iron, increasing the labile iron pool and upregulating ferritin. A new genetically encoded fluorescent reporter showed an inverse correlation between intracellular polyamine levels and redox-active iron in single living cells, repositioning polyamines as regulators of iron homeostasis with implications for ferroptosis-linked disease states.

Dynamic epidemiological changes of hand, foot, and mouth disease and real-world effectiveness of EV-A71 vaccination: A case study in Shanghai (2009-2023)

EV-A71 vaccination in Shanghai's Minhang district cut HFMD incidence by 56.7%, case-severity by 95.2%, and fatality by 100.0% between 2009 and 2023, using 73,160 reported cases in a test-negative design. Two-dose vaccine effectiveness against EV-A71-associated HFMD and herpangina was 89.9%, with no cross-protection against other serotypes, which shifted from EV-A71 and CV-A16 to CV-A6 and CV-A10. The serotype shift supports multivalent vaccine development.

Antifungal therapy improves microbiome dynamics in inflammatory bowel disease.

Fluconazole, but not nystatin, reduced intestinal Candida burden and reshaped gut fungal-community composition in 53 patients with mild-to-moderate ulcerative colitis or Crohn's disease. The gastrointestinal and oral fluconazole fungal targeting arm (n = 35) showed increased bacterial diversity, expansion of short-chain fatty-acid-producing taxa, restored anti-inflammatory metabolites, and improved disease activity indices with decreased risk of progression over 8 weeks. The work establishes a framework for antifungal cotherapy in fungal-associated IBD.

Caloric restriction modulates genome-wide somatic mutation in mice

Caloric restriction reduces the accumulation of somatic mutations across the genome during aging in mice. Grońska-Pęski et al. measured mutation burdens genome-wide in mice under caloric restriction and found reduced mutation loads across several tissues and cell types. The work establishes that diet can alter genome-wide mutation accumulation during aging.

Impaired liver-muscle lactate metabolism causes sarcopenia via lactic acidosis in skeletal muscle in mice.

Impaired lactate processing in the liver drives lactic acidosis in skeletal muscle, and this emerges as a fundamental cause of sarcopenia in aged mice. Systemic lactate tolerance fell in aged mice, and lactic acidosis lowered nicotinamide adenine dinucleotide levels, disrupting skeletal muscle function. Pharmacological activation of hypoxia-inducible factor or liver-specific activation of HIF1α restored lactate tolerance, resolved muscle lactic acidosis, and improved sarcopenia.

Structural basis of a SWI/SNF-associated transcription pre-initiation complex.

SWI/SNF, long considered an independent chromatin remodeler, is instead an integral component of the transcription machinery: it associates with an RNA polymerase II pre-initiation complex, Mediator and a nucleosome, dimerizing into a 7-megadalton assembly. High-resolution cryo-EM structures of PIC-Mediator-SWI/SNF and PIC-Mediator-SWI/SNF-nucleosome complexes show SWI/SNF stabilizes PIC organization and remodels the +1 nucleosome. The model proposes coordinated action of two DNA translocases, Snf2 and Ssl2 of TFIIH, during initiation.

Kinetic assembly tuning NIR-II cyanine aggregated states: Remarkable photothermal-mediated sustained retention in orthotopic hepatoma.

A kinetic assembly method encapsulates the NIR-II dye DQP and protein in polymers, tuning the dye's aggregated state so monomers carry NIR-II fluorescence while aggregates drive photothermal conversion under single-wavelength excitation. The aggregated state's π-π stacking produces heat that triggers charge reduction and size enlargement of DQPNPs, increasing intracellular tumor retention in orthotopic hepatoma and improving both fluorescence intensity and photothermal efficacy. The work decouples imaging from therapy within one dye platform, easing the fluorescence quenching problem of conventional photothermal nanotheranostics.

Distinct ocular microvascular alterations in Alzheimer's disease and cerebral small vessel disease.

Alzheimer's disease (AD) and cerebral small vessel disease (CSVD) show distinct ocular microvascular patterns on optical coherence tomography angiography. Among 650 participants, CSVD patients had a higher burden of retinal ischemic perivascular lesions (RIPLs) than AD patients, while AD patients showed reduced choriocapillaris density, which correlated with elevated phosphorylated tau 181 (p = 0.001). Machine learning models using OCTA metrics differentiated AD from CSVD with an area under the curve of 0.86, supporting OCTA-derived measures as potential non-invasive biomarkers.
