FDA Approves AbbVie's Tavapadon for Parkinson's Disease

The FDA has approved AbbVie's tavapadon, a next-generation dopamine therapy for Parkinson's disease designed to deliver symptom relief comparable to earlier dopamine-based treatments with fewer side effects. Details on dosing, launch timing, and pricing were not disclosed, but the approval gives clinicians a new option in a therapeutic class where tolerability has long constrained care. The decision headlined a day that also brought a rare Trump administration move to claw back nearly $1 billion in congressionally approved spending, mostly HHS programs serving refugees and unaccompanied minors, alongside new human evidence linking the gut-derived metabolite indole-3-propionic acid to type 2 diabetes risk and glycemic variability.

Trump administration uses rare authority to claw back nearly $1B in spending approved by Congress

President Donald Trump is canceling nearly $1 billion in spending approved by Congress, mostly HHS programs serving refugees and unaccompanied minors, using a contested "pocket rescission" announced with five days left in the fiscal year. The cuts also include $70 million for international education programs, $28 million for HHS research grants, and $10 million from a minority business development program. Congress has 45 days to review the proposal, and Sen. Susan Collins has vowed to challenge what she calls illegal impoundment.

Plasma indole-3-propionic acid is a gut-derived metabolite and is associated with type 2 diabetes and cardiometabolic outcomes: Evidence from a human antibiotic intervention

Plasma indole-3-propionic acid (IPA) is lower in individuals with type 2 diabetes than in healthy controls, and higher pre-antibiotic levels associate with reduced T2D odds. In both groups, 7-day broad-spectrum antibiotic treatment reduced IPA to near-undetectable levels, confirming its predominantly microbial origin. IPA also showed a novel inverse correlation with glycemic variability and linked to gut microbiota beta diversity rather than known IPA-producing taxa alone.

Plasma indole-3-propionic acid is a gut-derived metabolite and is associated with type 2 diabetes and cardiometabolic outcomes: Evidence from a human antibiotic intervention

Plasma indole-3-propionic acid (IPA) is lower in people with type 2 diabetes than in healthy controls and higher levels are associated with reduced T2D odds. A 7-day broad-spectrum antibiotic intervention, which suppressed gut microbiota in individuals given histidine supplementation, cut IPA to near-undetectable levels in both groups, confirming its microbial origin. IPA also showed a novel inverse correlation with glycemic variability, and was associated with gut microbiota beta diversity rather than any known IPA-producing taxa.

Assessing the consistency of overall survival extrapolations in Swedish health technology assessment reports of oncology treatments using extended follow-up data

Overall survival extrapolations in Swedish TLV oncology appraisals were frequently conservative: reapplying original assumptions to longer follow-up data raised the average area under the curve of extrapolated OS by 8.8% (95% CI 5.2% to 12.6%) under TLV assumptions and by 10.9% (95% CI 6.9% to 15.1%) under manufacturer assumptions. The findings argue for revisiting survival model fit and treatment-waning assumptions when longer-term data become available.

Assessing the consistency of overall survival extrapolations in Swedish health technology assessment reports of oncology treatments using extended follow-up data

Overall survival extrapolations in Swedish TLV oncology appraisals from 2009 to 2019 were often conservative for the active arm. Reapplying the original extrapolation assumptions to longer follow-up data (average 54.2 months versus 28.1 months originally) raised the average area under the survival curve by 8.8% under TLV assumptions and 10.9% under manufacturer assumptions, with material increases above 10% in roughly one-third of cases. The findings argue for revisiting survival model fit and assumptions as longer-term data emerge.

Discrepancies between ChIP-seq and CUT&Tag histone mark profiles are explained by GC content and chromatin accessibility

Systematic comparison of ChIP-seq and CUT&Tag histone mark profiles in K562 and MCF-7 cells shows that many disagreements between the assays track GC content and chromatin accessibility. CUT&Tag loses H3K36me3 signal in GC-poor active gene bodies and H3K27me3 signal in GC-poor Polycomb-repressed domains, and promoters with discrepant H3K4me3 and H3K27ac signal differ systematically in GC content and DNase-seq and ATAC-seq signal. Studies profiling histone modification intensity and structure should account for these biases.

SHERLOCK: Structured representation learning and causal inference of downstream perturbation effects

SHERLOCK is an interpretable deep generative framework that represents genetic, pharmacological, and experimental perturbations as structured interventions on a latent baseline cellular state within a structural causal model. It learns correlated and sparse perturbation representations, organizes perturbations by shared transcriptional responses, and supports counterfactual estimation of downstream transcriptional effects, condition-dependent response analysis, and compositional modeling of combinatorial perturbations. Across genome-scale CRISPR, chemical, and spatial perturbation datasets, it recovers perturbation relationships concordant with known pathways and pharmacological properties and predicts held-out perturbation combinations.
