Engineered genomic attachment sites for site-specific recombinases enable high-efficiency integration in plants and human cells
Rationally designed chimeric attB sites, built by recombining favorable sequence motifs from natural Bxb1 recognition sites, yield hyperactive attachment sites that boost recombinase-mediated targeted integration. From 144 designed attB variants, all recognized by Bxb1, the authors isolated variants driving efficient integration in human cells and rice, including a CD19-targeted CAR and an OTC cassette, with specificity confirmed by NGS and Nanopore sequencing and Mendelian inheritance in T1 rice lines.
A multi-functional oral small molecule targeting energy and lipid metabolism to treat obesity and related metabolic disorders.
An orally bioavailable small molecule, 5-tetradecyloxy-2-furoic acid (TOFA), alleviated obesity, abnormal glucose homeostasis and fatty liver-related disease without affecting food intake or muscle mass. TOFA inhibits acetyl-CoA carboxylases 1 and 2 and activates PPARalpha/delta, raising energy expenditure and rebalancing lipid synthesis. It also acted more than additively with the incretin analogs semaglutide and tirzepatide to improve obesity, dyslipidemia and insulin resistance.
Antigen-experienced airway CD8+ effector memory T cells promote a detour pathway for mycobacterial killing in the spleen.
Repeated airway exposure to Bacille Calmette-Guérin generates fewer but more effective CD8+ effector memory T cells, which clear Mycobacterium tuberculosis faster by trafficking to the spleen and recruiting dendritic cells that accelerate bacterial translocation from the lung. Transfer of a threshold number of these cells significantly reduced Mtb burden in lung and spleen, expediting bacterial killing by at least 15 days. The work identifies a spleen-centered protective mechanism relevant to TB vaccine design.
Cell-type-specific eQTLs underlie the genetic architecture of complex traits.
Cell-type-specific eQTLs, mapped with a variance component model applied to population-scale single-cell RNA-seq of peripheral blood mononuclear cells from the OneK1K cohort, enrich for complex trait heritability, unlike cell-type-shared eQTLs. Trans eQTLs were mostly cell-type-specific (60%) while cis eQTLs were mostly shared (30% specific). Findings, replicated in a second dataset across ancestries, explain partly why known eQTLs are depleted in gene regulatory effects on complex traits.
Responsiveness of epigenetic aging biomarkers to longevity interventions in humans.
A harmonized database of 51 longitudinal interventional studies, TranslAGE, was curated to test the responsiveness of 16 epigenetic clocks and 94 other DNA methylation biomarkers to longevity interventions. Clocks trained to predict mortality or pace of aging responded most strongly and consistently, while pharmacological and lifestyle interventions drove the strongest DNAm biomarker responses. The results guide the choice of interventions, biomarkers, study duration, population and sample size in future trials aiming to use DNAm biomarkers as surrogate aging endpoints.
An encyclopedia of human enhancer-gene regulatory interactions.
A resource of more than 92 million enhancer-gene regulatory interactions across 1,458 biosamples covering 369 cell types and tissues was built by integrating predictive models, chromatin states, three-dimensional contacts and ENCODE genetic perturbations. Benchmarking used 10,356 CRISPR-tested element-gene pairs, over 30,000 fine-mapped eQTLs and 569 fine-mapped GWAS variants, and produced ENCODE-rE2G, a model that improves linking noncoding variants to target genes and cell types in common complex diseases.
Cuproptosis-immunity crosstalk informs strategy to overcome immunotherapy resistance.
CD8+ T cell antitumor immunity makes tumor cells more susceptible to cuproptosis: T cell-derived IFN-γ drives FDX1 transcription via the STAT1-IRF1 axis, while cuproptotic tumor cells act as immunogenic cell death that activates dendritic cells. Combining a cuproptosis inducer with anti-PD-L1 therapy overcame PD-L1 therapy resistance across multiple preclinical models. The work links antitumor immunity to cuproptosis and identifies a way to counter immunotherapy resistance.
Modular in vivo antibody-ADC click to reverse drug resistance in tumours.
Sequentially administered antibodies and ADCs bearing trans-cyclooctene and tetrazine moieties ligate in vivo to form functional antibody-ADC click constructs, removing the dependence on a single target antigen. In preclinical models of HER2 and EGFR co-expression, including tumours with low, ultralow, negative or heterogeneous HER2 expression and tumours resistant to or ineligible for conventional HER2-directed ADCs, the approach showed improved antitumour activity over standard ADC monotherapy and over antibody plus ADC combinations. The platform requires no extensive antibody re-engineering and extends to other receptor pairs.
Oxygen-responsive bacterial glycosphingolipid links symbiont fitness and immune development in neonatal host.
Bacteroides fragilis colonizes the neonatal gut using the membrane glycolipid alpha-galactosylceramide (BfaGC), whose biosynthesis is selectively required during early life, when transient oxygenation creates a bottleneck for strict anaerobes. BfaGC reduces membrane proton permeability, sustaining the proton-motive force for aerobic respiration, while also calibrating neonatal natural killer T cell development. Enterotoxigenic strains exploit the same mechanism for niche expansion, and other gut Bacteroidales use a different sphingolipid subclass.
Excessive epithelial mechanosensation drives nociceptive innervation and chronic bladder pain via the PIEZO1-SLC7A11-glutamate axis.
Recurrent uropathogenic E. coli infection upregulates the mechanosensitive channel PIEZO1 in bladder epithelial cells through IL-6-dependent signaling, and this overexpression amplifies mechanotransduction-induced reactive oxygen species. The resulting antioxidant response via the cystine-glutamate antiporter SLC7A11 causes extracellular glutamate accumulation, driving sprouting and hyperinnervation of peptidergic nociceptive C fibers into the epithelium and producing persistent pain-like states. The work identifies the PIEZO1-SLC7A11-glutamate axis and the neuroepithelial interface as therapeutic targets for infection-induced chronic pain.
Isthmin 2 could determine the early origins of preeclampsia and fetal growth restriction
Low maternal serum isthmin 2 (ISM2) levels in early pregnancy predict preeclampsia and fetal growth restriction, two major causes of fetal morbidity and mortality worldwide. ISM2 proved essential for trophoblast invasion of the uterus, the process impaired in both conditions, pointing to a circulating factor that links placental development to disease onset.
Cyclin E-CDK2 regulates cancer cell transcriptional program and response to immunotherapy through BRD4
Cyclin E-CDK2 phosphorylates BRD4 and controls its chromatin occupancy, a transcriptional role independent of the cell cycle. Cyclin E overexpression represses interferon-stimulated gene expression in cancer cells through BRD4 and promotes resistance to immunotherapy, while inhibiting CDK2 activity enhances responses to immune checkpoint blockade. The work positions CDK2 as a targetable driver of checkpoint blockade response.
Late-life semaglutide treatment slows ageing and extends lifespan in female mice.
Semaglutide treatment of 20-month-old female C57BL/6 mice for 3 months improved physiological function, attenuated hallmarks of ageing and extended lifespan. In a longitudinal comparison with matched calorie restriction, semaglutide preserved baseline function and outperformed calorie restriction in exploratory drive, spatial memory and glucose control. The work frames GLP-1R activation as a calorie restriction mimetic acting partly through mechanisms beyond reduced food intake.
Dual antithrombotic therapy using potent antiplatelet inhibitors in atrial fibrillation and acute coronary syndrome: a randomized controlled trial.
In the EPIDAURUS open-label randomized trial in patients with atrial fibrillation and acute coronary syndrome, 1 month of a DOAC plus a potent P2Y12 inhibitor (prasugrel or ticagrelor) was compared with DOAC plus clopidogrel and in-hospital aspirin. The trial stopped early after 602 of 1,474 planned patients on Data and Safety Monitoring Board safety concerns. Exploratory analyses showed higher BARC type ≥2 and ≥3 bleeding with potent inhibitors and no clear reduction in ischemic events.
Antibody framework engineering enables augmented cytokine receptor signaling capacities of bispecific single domain antibody-based IL-21 mimetics
Bispecific single-domain antibody-Fc fusions targeting IL-21R and IL-2Rγ act as IL-21 mimetics, initially triggering weaker STAT3 phosphorylation than the wild-type cytokine in NK-92 cells and PBMCs. Framework mutations that rigidify VHH:VHH interactions, plus reformatting into an IgG-like scaffold, raised agonism to levels comparable to wild-type IL-21, including pSTAT3 induction and Granzyme B expression in cytotoxic T cells. The work shows paratope geometry, not valency, governs receptor agonism.

Metagenome-scale modeling to assess microbiome metabolic complementarity for precision microbiota transplantation therapies
Metagenome-scale metabolic modeling quantifies metabolic niche complementarity between donor and recipient microbiomes and predicts post-FMT community composition. Donor genomes with metabolic flux profiles more dissimilar to the recipient community colonized at significantly higher rates in a murine FMT model, and the metric predicted post-FMT composition in a human IBS trial. A total of 2,548 in silico FMT simulations between IBS-D/M patients and OpenBiome donors identified super-donors with high diversity and cross-feeding-dominated networks.
Design of aptamer peptides for immunomodulation
Two peptide aptamers built on the sunflower trypsin inhibitor-1 (SFTI-1) template block the CD2-CD58 co-stimulatory interaction and resist enzymatic degradation. SFTI-FGUA carries a side-chain guanidine group on phenylalanine and SFTI-DMY a meta dimethyl group on tyrosine, substitutions that restrict conformational flexibility and improve solubility. In cell adhesion inhibition assays both peptides significantly inhibited cell adhesion and showed a predominant conformation in solution.

Atypical p38 kinase signaling and pathological impact on human health
A review by Royer, Grimsey and colleagues in the Biochemical Journal synthesizes evidence that p38 MAPKs, especially the p38α isoform, can be activated through atypical mechanisms independent of the upstream kinases MKK3 and MKK6. It compares interactors including TAB1, ZAP-70, the hepatitis C virus core protein and Toxoplasma gondii GRA24, and links this activation mode to cardiac ischemia, infection, inflammation and vascular dysfunction. The authors frame atypical p38 signaling as a distinct regulatory paradigm within MAPK signaling.
