NIK 稳定化重塑胰腺走向慢性胰腺炎与胰腺脂肪变性
NIK stabilization reprograms the pancreas toward chronic pancreatitis and pancreatic lipomatosis
This study used a pancreas-specific NIK stabilization mouse model to show that sustained non-canonical NF-κB signaling drives chronic pancreatitis and pancreatic lipomatosis. NIK stabilization caused rapid, fully penetrant pancreatic remodeling with acinar cell death by the end of the first week of age, followed by progressive loss of acinar identity, ADM formation, and identification of GPA33 as a novel ADM surface marker via spatial transcriptomics. ADM lesions subsequently underwent apoptosis, leading to parenchymal loss, organ atrophy, and extensive adipose replacement, all fully rescued by RelB deletion.
为什么重要
The work defines a RelB-dependent pathway linking sustained NIK stabilization to chronic pancreatitis and lipomatosis, addressing the poorly understood mechanisms driving pancreatic lipomatosis. The model provides a tool for studying lipomatosis development and exploring therapeutic approaches, while GPA33 identification offers a potential marker for tracking metaplastic progression. These findings clarify the role of non-canonical NF-κB signaling in pancreatic remodeling, which has been context-dependent in prior studies.
还缺什么
The study is a preprint and has not undergone peer review. Key translational questions remain, including whether the NIK-RelB axis operates similarly in human chronic pancreatitis and lipomatosis, whether GPA33 is detectable and functional in human ADM lesions, and whether pharmacologic NIK or RelB inhibition can reverse established disease. Therapeutic validation and human relevance await confirmation.
