@Aiims1742 At fixed daraxonrasib 100 mg, increasing vopimetostat from 200 to 250 mg yielded 3 DLTs in 2/4 patients. That makes dose central to the crosstalk hypothesis: tolerability at one dose does not yet show PRMT5-mediated protection of normal tissue.

Mechanism wise there has got to be a bi-directional crosstalk between wild type RAS and PRMT5 First Daraxonrasib has greater ORR than Zoldonrasib per press release, and second: “and partly because targeting PRMT5 may attenuate some of Rasonque’s toxicities”
