Weekly cisplatin fails to show fewer serious side effects in p16-negative head and neck cancer

Weekly cisplatin at 40 mg/m2 with 70 Gy radiation did not reduce serious acute toxicity versus 100 mg/m2 every three weeks in 234 patients with p16-negative locally advanced head and neck squamous cell carcinoma in the phase II NRG-HN009 trial. The mean T-score ratio was 1.11 (p=0.77), with T-scores of 2.23 versus 2.48, and six-month locoregional failure was 4.6% versus 9.6%. The cohort did not advance to phase III, leaving the every-three-week schedule unchallenged on toxicity grounds.

Why it matters: It removes the assumption that weekly cisplatin is the gentler option for p16-negative head and neck cancer.
