Genetic and environmental factors shape DNA methylation linked to ADHD in childhood
Start here
A preprint posted to bioRxiv examines how genetic variants and environmental exposures relate to DNA methylation at ADHD-associated CpG sites across childhood and adolescence. In a longitudinal cohort of children aged 9.5 to 11.9, 19 methylation quantitative trait loci (meQTLs) explained up to 40% of methylation variance at five ADHD-associated CpG sites. LASSO regression in 126 children identified 17 environmental predictors of methylation. Household income and school quality of life were associated with declining methylation at specific sites over time, and income showed divergent effects by clinical course: protective in children whose ADHD remitted, and the opposite in those whose ADHD persisted. The work links genetic and environmental influences to epigenetic variation relevant to ADHD trajectories, with further validation still ahead given its preprint status.
