In the phase 2a GDFATHER-NEO trial, neoadjuvant visugromab plus nivolumab in MIBC yielded a pCR of 33% versus 8% with nivolumab plus placebo. The study enrolled N=31 newly diagnosed MIBC patients ineligible for or declining cisplatin; 95% CIs for pCR were 12-62% and 0-36%. After three cycles, major pathologic response rates were 60% versus 23%, and RECIST v1.1 radiographic overall response was 60% versus 15%.
