Neoadjuvant nivolumab with or without visugromab in muscle-invasive bladder cancer: the phase 2a GDFATHER-NEO trial.

In brief

In the phase 2a GDFATHER-NEO trial, neoadjuvant visugromab plus nivolumab in muscle-invasive bladder cancer (MIBC) yielded a pCR of 33% versus 8% for nivolumab plus placebo. The study enrolled N=31 patients with newly diagnosed MIBC who were ineligible for or declined cisplatin; 95% CIs for pCR were 12-62% and 0-36%. After three cycles, major pathologic response was 60% versus 23%, and RECIST v1.1 radiographic overall response rate was 60% versus 15%.

Why it matters

A nonrandomized exploratory trial in 31 patients with a nivolumab plus placebo control allows comparison of pCR and major pathologic response with added visugromab.
