Abstract CT011: Genomic HER2 amplification predicts for complete clinical response in a phase II study of induction tucatinib and trastuzumab combined with chemotherapy in locally advanced rectal adenocarcinoma (Presented at #AACR26 )
https://t.co/JaBPDADpqy
This prospective single-arm study evaluated HER2-targeted 🧬 therapy in stage II–III HER2-positive rectal cancer. Patients received trastuzumab 🧪 and tucatinib with chemotherapy, achieving a 78% initial response and 75% overall response rate.
Complete clinical response occurred in select patients, particularly with strong HER2 expression, enabling non-operative management. With no metastatic recurrences at 26 months, results suggest HER2 amplification may predict response and support organ-preserving strategies in carefully selected patients.
