New results from the phase 2 FOURLIGHT-1 study show promising activity with atirmociclib + fulvestrant in patients with HR+/HER2− metastatic breast cancer who had progressed on prior CDK4/6 inhibitor therapy.

PFS: 9.4 vs 6.6 months (HR 0.60; P=0.0007)
ORR: 19.3% vs 11.3%
DOR: 14.8 vs 9.3 months
CBR: 61.9% vs 46.0%
OS: data not yet mature
The results support further evaluation of selective CDK4 inhibition, including in earlier treatment lines.
