Shallow nanopore sequencing method offers rapid QC for chromatin assay libraries
Start here
Researchers have introduced ONT-LibQC, a quality-control workflow for CUT&RUN and Micro-C chromatin assay libraries. Before committing to deep Illumina sequencing, the method reconstructs finished library molecules from roughly 3,000 shallow Oxford Nanopore reads, automatically identifying adapters and indexes and assessing fragment size distributions and enrichment of biological signal. The approach is designed to catch poor-quality libraries early, avoiding wasted deep sequencing runs. The work was posted as a preprint in October 2026 and has not yet undergone peer review. Open questions include how the method performs across diverse library types and assay conditions, and whether labs beyond the developers will adopt it as standard practice.
