Cytoplasmic localization of 5hmC reveals RNA hydroxymethylation in breast cancer
In brief
Wen et al. report in a bioRxiv preprint a quantifiable cytoplasmic 5hmC signal in breast cancer cell lines and human breast cancer tissue. RNase-sensitive, RNA-specific ELISA confirmed the signal reflects total RNA and mRNA hydroxymethylation, enriched in triple-negative (ER-/PR-/HER2-) breast cancer and grade 3 tumors, and correlated with increased cytoplasmic TET2/TET3.
Why it matters
Tissue signal assessed by digital pathology and confirmed as total RNA and mRNA hydroxymethylation by RNase-sensitive, RNA-specific ELISA, providing a method reference for similar assays.
