Combination therapy reveals potential vulnerability in aggressive osteosarcoma

In brief

A St. Jude Children's Research Hospital team found that combining EP300 degradation with AKT inhibition produced a synergistic effect in patient-derived mouse xenograft models, slowing tumor growth and prolonging survival. Most osteosarcoma samples sensitive to EP300 degradation carried AKT pathway-activating mutations, and sensitive tumors had a lower overall protein synthesis rate than resistant tumors; these mutations may serve as sensitivity biomarkers for the combination.

Why it matters

Links AKT pathway-activating mutations to EP300 degradation sensitivity in a subset of osteosarcomas, supporting biomarker selection for combination therapy.
