MCT6 is an intestinal Lac-Phe exporter required for metformin-associated weight loss.

In brief

A study identifies MCT6/SLC16A5 as the intestinal Lac-Phe exporter required for metformin-associated weight loss. MCT6 transports Lac-Phe in vitro and mediates cellular efflux; its loss disrupts circulating Lac-Phe levels, especially after metformin treatment or high-intensity exercise. High-fat-diet-fed mice with global or intestinal epithelial-specific MCT6 deletion resist metformin-associated weight loss, and exogenous Lac-Phe restores the weight phenotype of MCT6-knockout mice.

Why it matters

Global and intestinal epithelial MCT6 knockout plus Lac-Phe rescue directly link the transport defect to metformin-associated weight loss.
