Lung neuroendocrine cancer induces immunosuppressive macrophages via serotonin to drive resistance to the T cell engager tarlatamab.

In brief

Lung neuroendocrine carcinomas with high DLL3 expression upregulate TPH1 to increase serotonin (5-HT), which induces immunosuppressive tumor-associated macrophages (TAMs) expressing HTR7. This impairs CD8+ T cell function and drives resistance to tarlatamab.

Why it matters

Links the NEC-derived 5-HT-HTR7 axis to TAM immunosuppression and tarlatamab resistance; deleting TPH1 in NECs or HTR7 in TAMs restores CD8+ T cell function.
