Proteome-wide C-degron activity profiling connects conditional regulation of the CTLH E3 ligase complex to ribosome biogenesis.
In brief
The study developed an expression screening strategy to assess how C-terminal capping affects protein stability, systematically identifying multiple C-degron substrates of the CTLH E3 ligase complex across the human proteome. Muskelin-mediated C-degron pathways were confirmed on ZMYND19, AAMP, and AEN: ZMYND19 degradation was attenuated by TNF-α and enhanced by mTOR inhibition, while AAMP participates in ribosome maturation through chaperone activity toward uL16.
Why it matters
The paper establishes a C-terminal screening strategy to systematically identify degron substrates and links CTLH-mediated conditional degradation to ribosome biogenesis.
