Regulation-by-Partition model predicts enhancer target gene selection
Start here
A research team has proposed the Regulation-by-Partition (RbP) model, which allocates an enhancer's regulatory output among candidate target promoters according to genomic distance, contact frequency, and intrinsic promoter features, in order to predict enhancer target gene selection in mammalian genomes. To test the model, the researchers used piggyBac-mediated SPIE-seq, analyzing 29,954 mapped insertions across 724 clonal lines and detecting enhancer-dependent expression changes at 18,440 enhancer-gene pairs. The work, posted as a preprint on bioRxiv in Genomics on October 24, 2026, offers a quantitative framework for how enhancers choose their targets, a long-standing open question in gene regulation.
