AKT blockade degrades YAP to overcome resistance to RAS(ON) inhibition in pancreatobiliary cancer
Start here
A preprint posted on bioRxiv in Cancer Biology reports that blocking AKT drives degradation of the YAP protein through an ESCRT-dependent lysosomal pathway. In pancreatobiliary cancer, tumors adaptively resist inhibitors of RAS(ON), and this study finds that adding AKT blockade removes a key escape mechanism by eliminating YAP. The work suggests a combination strategy of RAS(ON) inhibition plus AKT inhibition for this cancer group. As a preprint, the findings have not yet undergone peer review, and questions of clinical translation, dosing and patient selection remain open.
