The phase 3 PRABITAS trial enrolled 970 patients with refractory metastatic colorectal cancer and tested a biweekly FTD/TPI plus bevacizumab regimen against the conventional regimen. The biweekly schedule was non-inferior for overall survival, with a median OS of 11.3 months versus 10.3 months (HR 0.93, 95% CI 0.80-1.08; non-inferiority P<0.0001). Superiority over the conventional regimen was not demonstrated. The biweekly regimen was also associated with fewer hematologic adverse events, suggesting a better tolerated option for this heavily pretreated population. Open questions include whether the efficacy and safety findings will translate into regulatory review or changes to standard practice in refractory metastatic colorectal cancer.
