Synthesis

The Phase 3 HERIZON-GEA-01 trial, run by BeiGene, tested zanidatamab plus tislelizumab and chemotherapy as first-line treatment for HER2-positive advanced gastroesophageal adenocarcinoma. At a second interim overall survival analysis, after a median follow-up of 35.3 months, the triple regimen achieved a median overall survival of 27.0 months versus 19.6 months in the control arm, with a hazard ratio of 0.65. The zanidatamab-plus-chemotherapy arm showed a median overall survival of 22.9 months (HR 0.76 versus control, P=0.0088). The difference between the tislelizumab-containing regimen and the zanidatamab-plus-chemotherapy arm did not reach statistical significance, leaving open the question of how much the PD-1 antibody adds.
