At ESMO 2026, three phase II studies examined treatment strategies after targeted therapy in BRAF V600E-mutant metastatic colorectal cancer. The ECLYPse trial met its primary endpoint, TRIDENTE demonstrated clinical activity, and BRICKET, which tested ctDNA-guided retreatment, recorded no objective responses. Together the trials map a mixed picture of options for patients whose disease has progressed on prior targeted therapy in this molecular subtype. Detailed results beyond the headline outcomes have not yet been reported, and questions remain open about which patients are most likely to benefit and how these strategies should be sequenced in practice.
