Spatial and single-nucleus maps chart aging and senescence across the human hippocampus
Start here
A preprint posted to bioRxiv Genomics maps how aging and cellular senescence unfold in post-mortem adult human hippocampi, combining spatial transcriptomics, single-nucleus transcriptomics and multiplex immunofluorescence imaging. The study finds that cell type, anatomical layer and sex jointly shape aging and senescence signatures. Synaptic genes of inhibitory neurons in the CA4 layer decline with age. In females, a microglial and neurovascular program that is highly expressed includes senescence-related genes such as CDKN1A/p21, and candidate senescence phenotypes defined by p16 and p21 also differ between groups. The work has appeared as a preprint and has not yet undergone peer review or journal publication.

A study finds that cell type, anatomical layer, and sex jointly shape aging and cellular senescence signatures in the post-mortem adult human hippocampus. Using spatial transcriptomics, single-nucleus transcriptomics, and multiplex immunofluorescence imaging (DOI 10.64898/2026.10.23.761418), the authors show that synaptic genes in CA4 inhibitory neurons decline with age. A microglial and neurovascular program highly expressed in females includes senescence-related genes such as CDKN1A/p21, and candidate senescence phenotypes defined by p16 and p21 also diverge.
