Multiomic characterization of microsatellite-stable colorectal cancer reveals distinct biology and targetable vulnerabilities.

In brief

A multiomic study using ~2,000 tumor samples reveals the distinct biology and targetable vulnerabilities of microsatellite-stable (MSS) colorectal cancer. The analysis covers 316 previously uncharacterized MSS CRCs, ~4,500 molecular profiles and 195,246 single cells or nuclei, and applies a multiomic-phenomic integration framework plus the machine-learning tool iPhenMap to identify replication stress, glutaminolysis-driven metabolic reprogramming and MYC-E2F transcriptional networks.

Why it matters

Screens candidate targets by multiomics, then validates them in patient-derived tumoroids and matched xenografts, giving a path from screening to functional testing of candidate combinations for MSS colorectal cancer.
