Cold activates a BH2 metabolic network promoting longevity and proteostasis

In brief

In C. elegans, cold drives BH2 synthesis in muscle via CAT-4; BH2 is reduced to BH4 in distal tissues, where PAH-1 uses it to synthesize tyrosine, extending lifespan and improving cross-tissue proteostasis. BH2 or tyrosine supplementation likewise suppresses aggregation of toxic proteins including polyQ74, TDP-43(A382T) and FUS(P525L) in HEK293 cells and ALS iPS-derived motor neurons.

Why it matters

The study shows cold extends lifespan and improves proteostasis via muscle BH2 synthesis and intertissue transport in worms and human cells, partly without cold.
