GRP75-dependent SIMPA mediates inter-organellar quality control.

In brief

Research shows that a GRP75-dependent SIMPA signalling pathway mediates quality control between mitochondria and the endoplasmic reticulum. Under proteostatic stress, damaged mitochondrial proteins are sequestered to form stress-induced mitochondrial protein aggregates (SIMPAs). Quantitative proteomics and correlative light-electron microscopy show that SIMPA activates the ER unfolded protein response in a spatially confined manner at mitochondria-ER contact sites or mitochondria-associated membranes to clear aggregates.

Why it matters

Quantitative proteomics and correlative light-electron microscopy localize SIMPA to mitochondria-ER contact sites, revealing a cross-organellar quality control mechanism.
