Lifestyle-induced regulatory T cell fragility primes adverse events after immune checkpoint blockade

In brief

Dietary and microbial alterations can increase susceptibility to irAEs after immune checkpoint blockade by promoting IFN-γ-driven Treg fragility and cytotoxic tissue-resident T cell responses. Disrupting IFN-γ signaling specifically in Tregs restores tissue homeostasis and reduces irAEs without compromising antitumor immunity, suggesting Treg stability is a potential therapeutic strategy for limiting checkpoint blockade toxicity.

Why it matters

Links lifestyle-related Treg fragility to irAEs and suggests targeting IFN-γ signaling to reduce adverse events while preserving antitumor immunity.
