Phase II ECLYPse: Encorafenib Plus Cetuximab Beyond Progression With FOLFIRI in BRAF V600E mCRC

At #ESMO26, Maria Alessandra Calegari presented results from the Phase II ECLYPse trial (788RO), evaluating encorafenib plus cetuximab with FOLFIRI in 25 patients with BRAF V600E-mutated metastatic colorectal cancer progressing on second-line encorafenib plus cetuximab.

Key findings:
🔹 6-month PFS: 66.7% (12/18 evaluable patients); Kaplan–Meier estimate 69.5% (95% CI 51.7–93.6%).
🔹 Median PFS: 7.1 months (95% CI 6.2–not estimable).
🔹 ORR: 15.0%; DCR: 80.0%.
🔹 Median OS: Not reached; 12-month OS rate was 83.7%.
🔹 Safety: Grade 3–4 treatment-related AEs occurred in 20% of patients, with no new safety signals.

Takeaway: ECLYPse met its primary endpoint, suggesting that continuing encorafenib plus cetuximab with FOLFIRI beyond progression may provide disease control. Further investigation is warranted given the small, single-arm design.
