Multimodal functional characterization of the LDLR noncoding landscape
In brief
Researchers used saturation CRISPR nuclease screening, CRISPR activation and inhibition screens, and a massively parallel reporter assay to systematically characterize noncoding cis-regulatory regions at the LDLR locus. Each method identified previously unrecognized regions affecting LDLR expression, but the CRE sets overlapped without being identical and also differed from computational predictions. DOI: 10.64898/2026.10.22.761041.
Why it matters
Multiple functional genomics methods each found previously unrecognized LDLR regulatory regions, with overlapping but distinct hit patterns.
