In vivo Gene Writing with engineered retrotransposons.
In brief
Researchers developed a computational pipeline to systematically identify and engineer non-LTR retrotransposons, producing Gene Writers that use trans-introduced RNA templates to insert reporter genes efficiently and stably in human cells. Gene Writers generated CD19 and CD20 CAR-T cells ex vivo with robust antitumor activity. Lipid nanoparticle-mediated RNA delivery enabled in vivo CAR-T cell generation and stable B cell depletion in humanized mice.
Why it matters
The paper presents the full path from computational identification and engineering of non-LTR retrotransposons to RNA delivery, with in vivo CAR-T generation and B cell depletion as application results.
