Computational design of metalloproteases.
In brief
Researchers used RFdiffusion to de novo design metalloproteases with water-activating zinc active sites. Of 135 designs, 36% catalysed hydrolysis at the targeted peptide bond; the best design accelerated peptide-bond hydrolysis by more than 10^8-fold, and four substitutions improving active-site preorganization raised this to 10^10-fold.
Why it matters
Demonstrates an RFdiffusion method for de novo design of zinc-active-site metalloproteases and the catalytic accelerations achievable, informing design of other enzyme classes.
