Metazoan lagging-strand DNA synthesis is distributive.
In brief
Metazoans complete lagging-strand DNA synthesis in a distributive manner, rapidly recruiting new Pol alpha and Pol delta for each Okazaki fragment rather than relying on stable polymerase docking. Using single-molecule imaging of active replication forks in Xenopus egg nuclear extracts, the study found that neither Pol alpha nor Pol delta is stably associated with the replisome, and leading- and lagging-strand synthesis are not intrinsically coupled.
Why it matters
Single-molecule imaging of replication forks in Xenopus nuclear extracts distinguishes stable polymerase docking from rapid turnover, providing experimental evidence for eukaryotic lagging-strand synthesis.
