A bioavailable GPX4 inhibitor for ferroptosis induction in vivo and tumor suppression.
In brief
A Cell study reports KY0418, an orally available covalent GPX4 inhibitor that specifically targets GPX4 SecCys46 to induce ferroptosis. In mouse models, it induces ferroptosis in vivo, remodels tissue metabolism, and eliminates circulating tumor cells to suppress metastasis. The inhibitor shows high plasma stability and broad tissue distribution; subcutaneous tumor models showed about 70% growth inhibition, and it consistently eradicated circulating tumor cells across multiple cancer models.
Why it matters
The study provides an orally available GPX4 inhibitor that induces ferroptosis in vivo and clears circulating tumor cells to suppress metastasis across models.
