Droplet-based organoid screens enable high-throughput identification of cancer vulnerabilities
In brief
The authors encapsulate tumor cells in hundred-micrometer-sized Matrigel droplets for organoid culture. A simple vortex step with standard laboratory equipment generates millions of uniform nanoliter compartments. The system can be combined with high-throughput drug screening, live-cell imaging, and pooled CRISPR screening.
Why it matters
Encapsulating organoids in hundred-micrometer Matrigel droplets enables high-throughput culture on standard equipment, easing replication of drug and CRISPR screens.
