Proteomic atlas of T cell exhaustion defines proteostatic collapse
In brief
Researchers built a multi-omic atlas of human T cell exhaustion and found that dysfunctional T cells undergo broad remodeling of protein turnover and ubiquitin regulation, changes not captured by transcriptomic analysis. The E3 ubiquitin ligase RNF166 regulates T cell persistence and anti-tumor function, pointing to proteostasis as a targetable axis for improving immunotherapy.
Why it matters
Proteomics reveals a proteostatic collapse mechanism of T cell exhaustion missed by transcriptomics and identifies RNF166 as a targetable node for improving immunotherapy.
