Firmonertinib in advanced non-small cell lung cancer with epidermal growth factor receptor P-loop and αC-helix compressing mutations.

In brief

The phase 1b FURTHER study (NCT05364073) evaluated first-line firmonertinib in metastatic non-small cell lung cancer with EGFR P-loop and αC-helix compressing (PACC) mutations. Confirmed objective response rates were 43% in the 160 mg arm (n=25) and 68% in the 240 mg arm (n=22), with median progression-free survival of 11.1 and 16.0 months, respectively.

Why it matters

First-line options for advanced NSCLC with EGFR PACC mutations are limited, and the response and progression-free survival figures allow direct comparison with other drugs targeting the same mutations.
