#ESMO26 | Abstract #LBA2 | VOLGA | Presented by @tompowles1 | @OncoAlert
✅ Primary endpoint (EFS): Durva+EV: HR 0.51 (95% CI 0.38–0.67; p<0.0001); Durva+Treme+EV: HR 0.61 (95% CI 0.47–0.80; p=0.0004)
✅ OS: Durva+EV: HR 0.58 (95% CI 0.40–0.83; p=0.0027); 2-year OS 78% vs 69%. Triplet did not demonstrate significant OS benefit.
✅ pCR: 45% vs 10% with control.
✅ Safety: G3/4 TRAEs 30% (Durva+EV) vs 42% (triplet). Adjuvant therapy completed in only 55% vs 42%, respectively. Toxicity with treme impedes ability to receive full course of therapy in some patients.
Practice implications: pCR less impressive than with KN-905. Suspect many will continue to use EV+P and more will feel comfortable omitting EV from the adjuvant phase of treatment after seeing this data, but response-adapted approaches for the adjuvant phase are the future, particularly for the patients who do not respond to neoadjuvant EV+ICI.
