Pharmacologic inhibition of OLIG2 by CT-179 disrupts mitotic progression and enhances standard-of-care chemoradiotherapy in glioblastoma

In brief

CT-179, a brain-penetrant small-molecule inhibitor of OLIG2, suppressed proliferation in all 11 patient-derived GBM cell lines (IC50 < 360 nM) and induced apoptosis through mitotic catastrophe, mitotic slippage, or a mixed response.

Why it matters

The paper links target engagement, cellular mechanism, and multiple patient-derived orthotopic models to build a preclinical chain from OLIG2 inhibition to extended survival.
