The landscape of gene loss and missense variation across the mammalian tree informs on gene essentiality

In brief

The authors developed two evolutionary constraint metrics, GISMO and GISMO-mis, across 462 mammalian species to complement assessments of gene essentiality. The two metrics provide complementary measures of gene essentiality and improve prioritization of human disease genes. The study was published in Nature Genetics, DOI 10.1038/s41588-026-02783-9, at https://www.nature.com/articles/s41588-026-02783-9.

Why it matters

Using gene loss and missense variation data from 462 mammals, GISMO and GISMO-mis complement gene essentiality assessment and improve human disease gene prioritization.
