Engineered human Argonaute 2 for targeted C-to-U RNA base editing.
In brief
Researchers engineered human Argonaute 2 (AGO2) into a modular RNA-targeting domain and fused it with a C-to-U deaminase to build the ALTER editing system. In HEK293T cells, ALTER achieved on average 1.9-fold higher C-to-U editing efficiency on mRNA targets than the Cas13 editors CURE and RESCUE.
Why it matters
Unlike Cas13-based RNA editing tools that depend on exogenous proteins, ALTER uses human AGO2 as its scaffold, offering comparable off-target and immunogenicity baselines.
