Engineered TYK2 dependence enables myeloid-selective IL-10 receptor agonism.
In brief
Researchers designed a tetravalent nanobody-based myeloid-selective IL-10 receptor agonist, the msIL-10R agonist, which reproduces IL-10's immunosuppressive effects on monocytes, macrophages and dendritic cells without activating T cells.
Why it matters
The work presents a receptor cross-linking geometry-based design for myeloid-selective cytokine signalling and validates a roughly 30-day sustained effect after a single dose in cynomolgus monkeys.
