Evidence for TROP2-directed antibody-drug conjugates (ADCs) has so far been established mainly in breast and ovarian cancer: published reviews show that sacituzumab govitecan and datopotamab deruxtecan demonstrated significant efficacy with acceptable safety in pretreated advanced triple-negative breast cancer and ovarian carcinoma, with trials reporting improvements in progression-free survival, overall survival and objective response rate versus standard therapies [11]. In endometrial cancer, options for advanced and recurrent disease were historically limited, and in recent years PD-1/PD-L1 inhibitors, PARP inhibitors, ADCs and hormonal therapy have reshaped the landscape [8]; sacituzumab tirumotecan (sac-TMT) has also been explored in this tumor type in combination with pembrolizumab for post-platinum pMMR advanced disease in the 1259P study [3]. Among the published literature provided, no phase III data for a TROP2 ADC in endometrial cancer were available.
This year's ESMO 2026 Presidential Symposium LBA11 filled that gap: the phase III TroFuse-005 evaluated sac-TMT versus physician's choice chemotherapy in previously treated recurrent/advanced endometrial cancer, with Domenica Lorusso among the discussants. The figures from the on-site posts can only be copied verbatim: median OS 19.0 vs 12.1 months (HR 0.68), median PFS 7.2 vs 3.9 months (HR 0.59), ORR 35.1% vs 17.3%, and nearly 7 months longer median survival; the speaker positioned it as a potential new standard after platinum plus immunotherapy. Another on-site view noted that TROP2 is expressed in most endometrial tumors, and that the ADC question is shifting from which target to which payload, and in what order.
The main-site news independently covered the same event, with OS figures consistent with the on-site posts: MSD/Kelun-Biotech announced global phase 3 data for sacituzumab tirumotecan in advanced or recurrent endometrial cancer after immunotherapy, with OS of 19.0 vs 12.1 months. What the news added beyond the posts were the full trial name TroFuse-005/ENGOT-en23/GOG-3095, registration number NCT06132958, and the 'after immunotherapy' population definition; the PFS, ORR and both HR values so far appear only in the on-site posts.
In terms of advancement, TroFuse-005 extends phase III evidence for TROP2 ADCs from breast and ovarian cancer [11] into post-immunotherapy endometrial cancer with an OS benefit, connecting with the broader evolution of immunotherapy and ADCs in this disease [8] and providing a monotherapy reference for combination strategies such as 1259P [3]. Limitations are equally clear: neither the on-site posts nor the news disclosed safety data, sample size, stratification factors, dMMR/pMMR subgroup results, follow-up duration or statistical significance details, and the composition of the control arm was not specified; all of these await confirmation in the formal abstract and full publication.
