Thymic innate-like T cells enforce CD8 tolerance through T-to-T antigen presentation
Start here
A lecture on the ImmuneZoom series described a mechanism in which thymic innate-like T cells, including NKT cells, act as long-term thymic residents expressing inflammatory mediators such as IFN-gamma, thereby forming a reservoir of inflammation-associated self-antigens. Using a GFP/IFN-gamma model antigen, Jedi TCR transgenic mice and thymic organoids, the speaker showed these cells trigger negative selection of CD8 T cells via direct T-to-T antigen presentation across roughly a 700-fold affinity range. As few as 0.1-0.55% of thymic innate-like T cells suffice to eliminate self-reactive CD8 T cells efficiently. Unpublished follow-up work identified V-delta-7 TCR recognition of the IL-17 receptor A chain as a ligand driving gamma-delta T cell selection; those findings have not yet been peer-reviewed.
