Organoid Studies Map Stem Cell Niche Signals and Secretory Cell Fate in Gut and Salivary Gland
Start here
An ImmuneZoom expert lecture presented work on human intestinal organoids and submandibular gland organoids. In intestinal organoids, LGR5+ stem cells at the crypt base differentiate into absorptive enterocytes and secretory lineages; human Paneth cells, unlike those in mice, may not supply Wnt to stem cells. Base-editing the IL-22 receptor abolished IL-22-induced Paneth differentiation, and knocking out ZNF800 sharply increased chromogranin A+ enteroendocrine cells while depleting goblet cells. Submandibular gland organoids retained the original gland's heterogeneity and supported HCoV-229E infection, offering a platform for studying transitions between homeostasis and inflammation. Open questions include how niche signals operate in human tissue and how these platforms translate to infection and inflammation research.
