SF3B1 Mutation Found to Generate Novel Immunogenic Peptides from Long Non-Coding RNAs
Start here
Long-read sequencing of full-length transcripts, analyzed with a new workflow called ALLR, shows that mutations in the splicing factor SF3B1 generate thousands of novel long non-coding RNAs with diverse alternative splicing patterns. Matched RNA and proteomic data indicate these lncRNAs are translated into peptides that show immunogenicity and affect cellular signalling. In primary chronic lymphocytic leukemia samples, 47 of the lncRNAs were detected and linked to adverse prognosis alongside known CLL risk factors. The findings, posted as a preprint, point to an unexpected source of antigenic material in SF3B1-mutant cancers, though independent validation and any translational application remain open questions.
